Safranin Color Uses in Biology:
- Counterstain in Gram Staining: Safranin is used as a counterstain in Gram staining, a method for classifying bacteria. After the primary stain (crystal violet) and a decolorizing step, Safranin stains the Gram-negative bacteria red or pink while Gram-positive bacteria remain violet.
- Staining Cell Nuclei: Safranin stains cell nuclei red due to its affinity for nucleic acids.
- Plant Tissue Staining: In botany, Safranin is used to stain lignified tissues, like xylem, and red, helping to differentiate between various plant structures.
Staining Protocol:
In a typical protocol (e.g., Gram staining), Safranin is applied as the final step after other staining and decolorization steps. It generally takes 30 seconds to 1 minute for the stain to penetrate the cells.
